In this interview, we discuss about this This year Nobel Prize in Chemistry has been given to the power of directed evolution mainly applied to enzymes and phage display. In this interview, we discuss about this technology with Prof. Dr. Andreas Plückthun, who has been working for many years on protein engineering and developed very useful systems such as ribosome display and the Designed Ankyrin Repeat Protein (DAPRin) technology. Directed evolution, pioneered by Arnold in the early 1990s, is a bioengineering method for creating new and better enzymes in the laboratory using the principles of evolution. Today, the method is used in hundreds of laboratories and companies that make everything from laundry detergents to biofuels to medicines.

Directed evolution of binding proteins is an efficient way to identify variants with high affinity and selectivity for a given target, and to map the sequence requirements for high-affinity and high selectivity protein interactions. Directed evolution of human antibodies leads to useful therapeutics. Figure 1. Evolution towards directed evolution There are three main steps in directed evolution: mutagenesis, selection/screening of the desired enzyme property, and decision of which parental genes will proceed to the next round of evolution. The experiments can be performed in vivo, inside cells (bacteria or yeast), or through in vitro transcription–translation systems. Directed evolution has been used by Arnold and others to make all manner of molecules that would otherwise require lengthy, expensive, and often environmentally unfriendly syntheses. The list of products made enzymatically includes the biofuel isobutyl alcohol and the antidiabetic drug sitagliptin.
